VC-SOP-0204 — GCL-4 Lethal Cultivar Containment & Handling
The mandatory procedure for entry, handling, and egress involving GCL-4 specimens — weaponized and aggressive cultivars whose toxin, predation, or dispersal profile is lethal to personnel. There is no acceptable shortcut in this document.
1. Purpose
To define the controlled method by which qualified personnel may approach, observe, sample, feed, or transfer GCL-4 specimens held in Bio-Vault containment spheres, such that containment integrity remains 100% and no personnel exposure occurs.
2. Scope
Applies exclusively to GCL-4-certified Containment & Bio-Vault personnel operating within the GCL-4 wing (Spheres S-41 through S-48). GCL-1/2 cultivation is governed by VC-SOP-0101; emergency breach response by VC-SOP-0401. This SOP supersedes all standing work instructions in the event of conflict.
3. Definitions
| Term | Definition |
|---|---|
| Containment Sphere | A graded, sealed isolation enclosure with independent atmosphere, negative pressure, and a UV/thermal sterilization ring. |
| Airlock | The two-door interlocked transfer vestibule between facility atmosphere and a sphere; both doors cannot open simultaneously. |
| Two-Person Rule | No GCL-4 task proceeds with fewer than two certified personnel: one Handler, one Safety Observer. |
| Negative Pressure | Sphere interior held below ambient (-25 to -40 Pa) so any leak flows inward. |
| Decon Sequence | The mandated multi-stage decontamination on egress (Section 8). |
4. Responsibilities
| Role | Responsibility |
|---|---|
| Containment Officer (Owner) | Authorizes each GCL-4 entry; verifies certification currency; owns this SOP. |
| Handler | Performs the in-sphere task. Never breaks visual contact with the specimen. |
| Safety Observer | Stationed at the airlock console; controls interlocks; holds abort authority; never enters. |
| CHLORA | Monitors sphere telemetry; arms auto-response; logs all door cycles and pressure data. |
| Watch Commander | Notified of every GCL-4 entry; standing Tier-3 readiness during the task. |
5. PPE Requirements
Per PPE Matrix VC-SOP-0301, GCL-4 entry mandates the full encapsulating ensemble. Entry is denied if any item fails inspection.
- Level-A encapsulating positive-pressure suit (integrity-tested < 24 h)
- Self-contained breathing apparatus, ≥ 45 min air, charged ≥ 90%
- Double nitrile + cut/puncture-resistant outer gloves, taped seams
- Chemical-splash boots over suit, taped
- Telemetry badge (heart rate, suit pressure, exposure dosimeter)
- Two-way MDF comms link, voice-confirmed with Safety Observer
6. Equipment & Pre-Entry
- Sphere telemetry green on the CHLORA console (pressure, atmosphere, specimen tracker)
- UV/thermal sterilization ring confirmed armed
- Long-reach manipulator tools (no bare-hand specimen contact, ever)
- Decon shower & neutralant reservoir charged
- Emergency severance / lockdown control verified within Handler reach
7. Procedure
7.1 Authorization & Two-Person Verification
- Handler and Safety Observer present GCL-4 certification to the Containment Officer; CHLORA verifies currency.
- Confirm the Two-Person Rule pairing in the console. If either certification has lapsed, entry is denied.
- Watch Commander acknowledges the entry; standing Tier-3 readiness is logged.
7.2 Airlock / Sphere Entry
- Safety Observer takes the airlock console; Handler dons PPE and passes a suit-integrity check.
- Observer opens the outer airlock door; Handler enters the vestibule; outer door seals.
- Vestibule runs a 30 s air-curtain purge; pressure steps down to sphere negative pressure (
-25 to -40 Pa). - Only when the outer door is confirmed sealed does the Observer release the inner door interlock. Both doors open simultaneously is a hard fault → abort.
- Handler enters the sphere, maintains continuous visual contact with the specimen, and voice-confirms position to the Observer.
7.3 In-Sphere Task
- Perform the authorized task (sampling, feeding, transfer) using long-reach manipulators only.
- Observer monitors specimen-tracker motion and Handler biometrics; calls any motion spike or dosimeter rise.
- Handler announces each discrete action before performing it. No unannounced movement near the specimen.
- Maximum continuous in-sphere time: 20 min or 60% air reserve, whichever comes first.
7.4 Egress
- Handler withdraws to the inner door and voice-confirms clear of the specimen.
- Observer cycles the inner door closed and confirms seal before the decon sequence begins.
7.5 Alarm Responses
| Alarm | Indication | Immediate Action |
|---|---|---|
| Pressure Drift | Sphere pressure rising toward ambient | Observer halts task; Handler holds position; CHLORA re-establishes negative pressure. Resume only when stable. |
| Specimen Motion | Tracker motion above species baseline | Handler freezes; Observer calls range; if specimen breaches manipulator standoff → abort to airlock. |
| Suit Breach | Suit pressure loss / dosimeter rise | Immediate egress; full decon; medical per VC-SOP-0307; report as Tier-4. |
| Containment Breach | Sphere integrity loss / dispersal detected | Execute VC-SOP-0401; CHLORA arms auto-response; facility lockdown. |
8. Decontamination Sequence (Egress)
- Gross knockdown: In the vestibule, Handler stands in the decon shower; neutralant misting for 90 s while rotating.
- Sterilization ring: Observer cycles the UV/thermal ring across the vestibule for one full pass.
- Outer-door release: Only after decon confirms complete does the Observer release the outer door; pressure equalizes to facility ambient.
- Doffing: Handler doffs PPE in strict reverse-don order at the doffing station; outer gloves first, SCBA last.
- Verification: Dosimeter read-out and suit-swab clear before the Handler exits the doffing station. Any positive → medical hold per VC-SOP-0307.
- Close-out: CHLORA archives the full door/pressure/biometric record; Containment Officer signs the entry closed.
9. Records
- GCL-4 entry authorization & two-person verification log
- Door-cycle, pressure, and biometric telemetry (CHLORA, retained 10 years)
- Decon completion & dosimeter clearance record
- Any alarm / abort / exposure report (cross-filed to VC-SOP-0401)
